چکیده

Some issues are considered which arise when template matching is used to identify cells in differential interference contrast (DIC) microscope images. An automatic method of counting and sizing the cells in such images is proposed and two examples given. Template matching is complex due to the nature of DIC images, namely the way in which light is captured across the image. Further complications arise due to different cell sizes and orientations. The method used compensates for these problems.

Towards automatic cell identification in DIC microscopy.

A general method is proposed for constructing templates of cells in differential interference contrast (DIC) microscopy. This takes account of the optics which generate DIC images, and is applicable to both transparent and semi-transparent cells of simple and complex shapes. Then, a template matching methodology is presented, which uses fast Fourier transforms to fit templates of a range of siz...

Multimodal Microscopy B Y Digital Image Processing

A matching algorithm is proposed for aligning microscope images obtained using diierent modalities, making use of cross-correlations of outputs from Prewitt's edge lter. Brightteld, phase contrast and differential interference contrast microscope images of algal and bacterial cells from an experimental, high-rate algal pond are used for illustration. The information content o f m ultimodal imag...

Evaluation of Similarity Measures for Template Matching

Image matching is a critical process in various photogrammetry, computer vision and remote sensing applications such as image registration, 3D model reconstruction, change detection, image fusion, pattern recognition, autonomous navigation, and digital elevation model (DEM) generation and orientation. The primary goal of the image matching process is to establish the correspondence between two ...

Reconstruction of optical pathlength distributions from images obtained by a wide-field differential interference contrast microscope.

An image processing algorithm is presented to reconstruct optical pathlength distributions from images of nonabsorbing weak phase objects, obtained by a differential interference contrast (DIC) microscope, equipped with a charge-coupled device camera. The method is demonstrated on DIC images of transparent latex spheres and unstained bovine spermatozoa. The images were obtained with a wide-fiel...

Real-time quantitative differential interference contrast (DIC) microscopy implemented via novel liquid crystal prisms

A phase shifting differential interference contrast (DIC) microscope, which provides quantitative phase information and is capable of imaging at video rates, has been constructed. Using a combination of phase shifting and bi-directional shear, the microscope captures a series of eight images which are then integrated in Fourier space. In the resultant image the intensity profile linearly maps t...
